People working for the Department of Mysteries are called Unspeakables.
This department is a major topic in Harry's fifth year. It deals with the mysteries of time, wisdom and (most likely) love.
To enter the DoM, one simply uses the lift found in the Atrium. Following a bare corridor will lead one to a black door. Upon opening said door, one finds oneself in a circular room with twelve plain, black, handleless doors all around it. The room is lit with candles burning blue. The floor is polished so that it almost looks like standing water. When the door to the circular room is closed, the walls of the room revolve rapidly, making it impossible to determine which door is which. The doors open easily with a push out.

The Locked Room
The Locked Room is a room inside the Department of Mysteries. It is on Level 9 of the Ministry of Magic.
This is the room that Harry Potter attempts to enter when he and fellow DA members (Ron Weasley, Hermione Granger, Ginny Weasley, Neville Longbottom and Luna Lovegood) rush to the Ministry of Magic. They do so because Harry believes (wrongly, as it transpires later) that Lord Voldemort has captured Sirius Black and is slowly torturing him to do something for him.
Entry into the Locked Room is not possible, even when Harry uses Sirius' knife to try unlocking the door. Later, when Harry learns that there exists a power that he has in abundance and Lord Voldemort has not, this power (presumably love or grief) is revealed, by Dumbledore, to be locked inside a room at all times. It could arguably be this room.
The Planet Room
A room that Ron Weasley, Ginny Weasley, and Luna Lovegood run into in order to escape the Death Eaters chasing after them, Harry Potter, Hermione Granger, and Neville Longbottom. It is a dark room filled with planets and (evidently) something similar to the anti-gravity mist used in the third task of the Triwizard Tournament.
